What is LINE And INST in Focusrite?

LINE is the primary signal level that travels through the Tip, Ring and Sleeve jack connection. They are commonly used when you connect any non-instrument devices to your audio interfaces like the outboard preamp or the processor or instruments that give output line level like keyboards, drum machines, etc.

INST is an important level signal which does travel through the Tip and Sleeve jack connection and it will need a preamp to raise it to Line level. The INST can be used when you connect a bass guitar, electric guitar or any other instrument directly to your audio interface.
